La chiesa attuale è frutto di una ricostruzione novecentesca di una primitiva chiesa fatta costruire tra il 1648 e il 1656 dalla Confraternita della Santissima Annunziata, che ne ha dato compito all’architetto Carlo Morello, lungo via Po, che era l’asse della nuova ampliazione della città verso il fiume.
All’inizio presentava una navata unica ed una facciata liscia, senza portici. Via via il fabbricato viene ampliato con l’aggiunta di coro e cappelle e si arricchisce dell’altare maggiore di Bernardo Vittone e di macchine processionali eseguite dai migliori artisti dell’epoca, come il Beaumont o Stefano Maria Clemente.
Il 1776 vedrà la facciata completata ma solo nell’ ottocento verranno aggiunti i portici, per uniformare la facciata alla conformazione della via.
Alcuni anni dopo la chiesa non versa in buone condizioni, inoltre è insufficiente ai bisogni della comunità servita dalla confraternita, pertanto nel 1913 ne viene deciso l’abbattimento.
La ricostruzione inizierà nel 1919 su progetto dell’ing. Giuseppe Gallo e si concluderà nel 1934, ma già nel 1928 era stata riaperta al culto e in seguito consacrata, mentre la facciata a portici reggente un loggiato a grandi arcate fu completata entro il ‘34.
La nuova chiesa è stata progettata secondo i canoni del barocco romano. Le principali strutture interne sono state ricollocate al loro posto.
Rispetto alla posizione originaria la nuova chiesa risulta spostata verso ovest per permettere l’apertura verso i portici di via sant’Ottavio.
La SS Annunziata subì alcuni danni durante i bombardamenti del 1942. Ha un portale in bronzo opera dello scultore G. Buzzi ed è famosa in tutta la città per il presepe meccanico.
